Bk-1361 (BK1361, cyclo-RlSKDK) is a cyclic peptide with RLsKDK (s= D-serine), which ACTS as an effective selective inhibitor of ADAM8 with IC50 of 120nM.No significant activity was shown for ADAM 9,10,12., 17, and 10 M mMP-2, -9, and -14;In the cell-based shedding assay, CD23 shedding was inhibited with an IC50 of 182nM, which reduced the migration/invasion of pancreatic cancer cells, decreased the activation of ERK1/2 and MMP, and caused changes in cell morphology.Reduced tumor load and transplanted pancreatic tumor cell metastasis, and provided Kras (G12D) driven improed indicators of clinical symptoms and survival in a PDAC mouse model.
