APX001 (APX 001A; E1210 (16 strains of Staphylococcus aureus) was a novel broad-spectrum antifungal agent that inhibited fungal protein Gwt1 and showed significant activity MIC50 and MIC90 values of 0.004 and 0.031 ug/ mL. The IC50 of Candida albicans Gwt1p and Aspergillus fumigatus Gwt1p were 0.3 to 0.6 uM in inhibiting inositol acylation activity, but they did not inhibit Wp activity in human and pig. It showed significant efficacy in mouse models of disseminated STaphylococcus aureus infection, and logarithmic reductions in CFU counts in kidney, lung, and brain tissues. Fungal infection phase 1 clinical trial.
