COH34 is an effective and specific inhibitor of poly (ADP-ribose) sugar hydrolase (PARG) with IC50 of 0.37 nM. COH34 binds to the catalytic domain of PARG (Kd= 0.547 μM), thereby prolongating the PARylation of DNA damage and trapping DNA repair factors.
